Why amniotic fluid?

Amniotic fluid is a complex and dynamic bioactive substance that provides mechanical support to the fetus during gestation. It contains a wide variety of carbohydrates, proteins, peptides, lipids, pyruvate, electrolytes, enzymes and hormones that create the ideal environment that protects, lubricates, and cushions during the development of human anatomies.1 Using the patent-pending Selectify process, Ascent combines amniotic fluid’s unique set of cells and cellular components in a single injection.

Ascent™ should be maintained in its original packaging and stored at ambient temperature (0°C to 38°C) until ready for use. When stored properly in their original packaging, Ascent™ allografts are shelf-stable for up to three (3) years.

It is made up of cells and cellular components retained from amniotic fluid. Amniotic fluid contains natural components, including proteins and growth factors, that protect the fetus as it grows and develops. Using a patent-pending method for processing amniotic fluid, Ascent captures these unique components in a single shelf-stable solution.
A range of factors including mechanical overloading, age, and genetics cause the body to degrade over time. These factors result in a complex series of biological events that cause inflammation and degeneration. In fluid environments within joints and tendons, this can cause cartilage breakdown, a change in synovial fluid composition, and microdisruptions. Ascent contains a unique array of natural biomolecular components, such as proteins, growth factors, cytokines, glycosaminoglycans, and enzymes, that counteract these changes by supplementing fluid environments to provide lubrication, cushioning, and protection, within joints, tendons and wound exudate.
The procedure for injecting Ascent is very similar to a simple steroid injection. It is quick, easy, and unlike many regenerative therapies, doesn’t require an invasive procedure. The injection site is cleaned, anesthetized and injected – all in less than 10 minutes.

Selectify is the proprietary method for processing Ascent. It begins with the selection of a variety of cell types called amniocytes. Once these desired cell types have been isolated, the remaining fluid is centrifuged to remove any undesirable components and purified using highly specialized filtration to retain a large group of key cellular components while also removing salts and other unnecessary molecules. The purified group of components includes proteins, growth factors, cytokines, and hyaluronic acid molecules that are beneficial for the reduction of inflammation and supplementation of nutrient deficient fluid environments.

The cells selected from the amniotic fluid during the first part of processing are recombined with the purified cellular components to create a concentrated solution that is preserved using lyophilization. The powder is then packaged and terminally sterilized.
